The Mushroom Castle appears as a location that can be visited in Super Mario RPG: Legend of the Seven Stars and its remake. It is home to Princess Peach, the Chancellor, Granny, the Vault Guard, and several other Mushroom People. The remake redesigns the castle slightly to change its rooftops from red to blue, reflecting its design on the map more clearly. The remake also includes a scene of Peach jumping out of the tower on-screen to rejoin Mario, something that had only been implied in the original game.

Some time after the events in Bowser's Keep, Mario is first escorted to the throne room by Toad, and the Chancellor is informed that the Princess has gone missing. Later, when Mario and Mallow return from Mushroom Way, they find Shymores exiting the castle's front door and most Mushroom People hiding in the Peach's Room. The first Star Piece is discovered after defeating Claymorton in the throne room, returning the castle to normal. Peach is later returned here after being rescued at Marrymore, though she escapes from her bedroom window using a parasol and joins Mario's party while Granny is disguised as her.

After obtaining the fifth Star Piece, Samus makes a cameo resting in the castle's guest room.
