The Duel of 100 is the Sammer Guy Tournament held in Sammer's Kingdom during the events of Super Paper Mario. Mario, Princess Peach, and Bowser enter it to receive the Pure Heart held by King Sammer. After twenty fights, The Void grows so large that Rolling Thwomp has to rush through the gates, plowing through the remaining Sammer Guys, and forcing the player to stop fighting for the moment until the end of the game. After the game is finished, however, the gang, now with Luigi, can reenter the tournament to fight all 100 Sammer Guys with unique names. Each Sammer Guy has its own introduction scene when the player enters the arena before starting the round.

There are 25 rounds per Chapter. The chapters serve only as checkpoints where Mario and his party can save, however; by using the Return Pipe they must start over from Chapter 6-1.

After defeating all 100 Sammer Guys, King Sammer rewards the team with seven Catch Cards. Each card contains a party member from Paper Mario: The Thousand-Year Door, which are Goombella, Koops, Flurrie, Yoshi, Vivian, Admiral Bobbery, and Ms. Mowz. When the player goes back to Sammer's Kingdom after beating the Duel of 100, King Sammer says he will banish the 100 Sammer Guys again and replace them with 100 cat robots, and thus the player cannot go back to fight the Sammer Guys again. This is a reference to Francis and his many cat robots.

There is an implied video game called Duel of 100: The Home Edition that is popular in Sammer's Kingdom. One Sammer Guy enjoys playing as himself.
