They act as the rulers of the [[Sprixie]]s, a race of fairies who play a major role in the story, and the princesses - which are green, yellow, blue, orange, purple, cyan, and red respectively - act as the game's damsel(s) in distress, replacing Peach in that role.

She first appears when [[Mario]], [[Luigi]], [[Princess Peach]] and [[Toad]] walk around the [[Clear Pipe]]. While they are investigating the Pipe, the Sprixie Princess pops out from the Clear Pipe and tells Mario and friends that the Sprixie Kingdom is in danger. As soon as she comes out, [[Bowser]] emerges from the pipe also. He then grabs the fairy princess and puts her into the bottle. The group enters the Clear Pipe to rescue her.

Moments after she emerges, however, [[Bowser]] comes up through the same pipe, captures the green Sprixie Princess in a bottle, and retreats down the pipe. The heroes quickly follow suite in order to rescue the besieged kingdom, and soon find that each sprixie princess is guarded in one of the first seven world's castles: defeating the boss of the castle will reveal a pipe that takes the party to a room containing both the [[Goal Pole]] and the imprisoned Sprixie.
 
The party eventually manages to rescue the last of the seven Sprixie Princesses after defeating Bowser in his purple cadillac-esque vehicle for the second time in [[World Castle]], seemingly the last stage of the game: however, Bowser promptly re-appears, stuffs all seven princesses into a larger bottle, and flees for [[World Bowser]], a Bowser-themed carnival that serves as the real final world of the game. Here, Bowser confronts the heroes after transforming into Meowser - essentially Cat Bowser - and is defeated, dropping the Sprixie Princesses in the process. The princesses now use their powers to undo all the harm done to their land before waving goodbye to the heroes as they return to the Mushroom Kingdom via a clear pipe.
