The player controls a ship at the bottom of the screen. It can move left and right, and when it moves off one side of the screen it reappears on the opposite side.  At the top of the screen are five panels equidistant from each other. Each of them has either a letter/exclamation mark or a question mark, the latter panels blink. The player's ship can shoot lasers upward by using {{button|GBA|A}}. When a laser hits a panel, it flips over and becomes a panel of the opposite state. To win, the player must have all panels display a letter/exclamation mark, causing them to spell a message. Once complete, the panels vanish and the letters/exclamation marks fly to the center of the screen while spacing themselves closer together. This makes them easier to read, as a visual flourish. Any remaining lasers vanish and the player loses the ability to shoot the second the task is complete.

Word Up is one of Dribble & Spitz's microgames in both the Sci-Fi folder in WarioWare, Inc.: Mega Microgame$! and the Fantasy level of the Mash League in WarioWare Gold. The name is derived from the 1986 song "Word Up!" by Cameo.
