True to its name, the tour introduced Mario and Donkey Kong Jr. in their Super Mario Kart sprite-based appearance as new drivers, the latter making his first playable appearance in the Mario Kart series since the aforementioned game. A new variant of SNES Rainbow Road, named RMX Rainbow Road 1, was added as the signature course of this tour. The drivers were announced in the Super Mario Bros. 35th Anniversary Direct on September 3, 2020.[1] Participating in the tour was one of the My Nintendo missions for the anniversary, with rewards including sweepstakes and a collectible pin set.[2] The menu's background featured the main intersection of RMX Rainbow Road 1 as the landmark for this tour.

The Super Mario Kart Tour reuses the Peach Cup's bonus challenge from the Tokyo Tour for this tour's King Boo Cup, the Koopa Troopa Cup's bonus challenge from the Ice Tour for this tour's Baby Mario Cup, and the Shy Guy Cup's bonus challenge from the Mario Bros. Tour for this tour's Shy Guy Cup.
Signature kart sets
In every cup, the driver which the cup is themed about is raised a tier on each track, making them a three- or two-item driver on that course. Up until the Ice Tour, a kart and a glider were also given a bonus for each race in the cup.
