Boss Sumo Bro is a Sumo Bro enlarged by Kamek's magic who first appears in New Super Mario Bros. U. He looks like a regular Sumo Bro, but is larger, has his eyes squinted, has a ring around his spike and appears to wear two tsuna around his shoulders, the ceremonial rope typically worn by yokozuna sumo wrestlers around their waists.

New Super Mario Bros. U / New Super Luigi U / New Super Mario Bros. U Deluxe
In New Super Mario Bros. U and its expansion, New Super Luigi U, the second tower of Rock-Candy Mines (called Screwtop Tower in New Super Mario Bros. U, and Sumo Bro's Spinning Tower in New Super Luigi U) appears to have no boss on top of it. However, when the player gets to the boss room, they find three floating platforms (with the floor under the side platforms slightly sunken in), four Note Blocks built in the center of the floor, and, on the other end of the room, a Sumo Bro. Magikoopa enters the room and enlarges the Sumo Bro to twice his original size, transforming him into an even stronger form called Boss Sumo Bro.
Like his original form, Boss Sumo Bro has a spiked head that makes it unsafe to jump on him, a black shell, and attacks by stomping the floor once, or, after a brief change in stance, twice in a row, with each stomping sending down sparks that spread electric shockwaves upon striking the floor. However, his eyes are now squinted much like how Sumo Bros squinted their eyes in Super Mario World as opposed to the beady eyes normal Sumo Bros have in this game (though, in Boss Sumo Bro's case, his eyes are permanently squinted in an angry expression), and he gains a pair of tsuna on his shoulders, as well as the ability to jump.
In battle, Boss Sumo Bro can leap to and from any of the three platforms in the room. Given his increased strength, his stomps cause earthquakes that will stun the player if they are standing on any solid ground at the time (much like the ground pounds of Sledge Bros, Morton Koopa Jr., and Roy Koopa), and his electric shockwaves travel across the entire floor rather than fizzling out after going a short distance. To attack him, the player must strike the platform he is standing on (the Note Blocks in the center of the floor, the flying squirrel form, Peachette, or the side platforms should be used when he is on the center platform, since it is higher than the other platforms). This would flip him on his shell, rendering him completely harmless even when touched and allowing the player to damage him by jumping on his exposed underside while he is stunned. As with Boom Boom, regardless of whether he is flipped or still actively moving around, the player can also pelt Boss Sumo Bro at any point in the fight (except for a short time when he is stunned after getting damaged) with six fireballs (the equivalent of one belly stomp), since Sumo Bros. are no longer fireproof in this game. After three stomps to the underside or 18 fireballs, Boss Sumo Bro falls off the stage, defeated. Like all tower bosses, he is briefly shown atop his tower once the player exits the level and returns to the map, before being toppled over and replaced with a flag.
Boss Sumo Bro is the only boss in New Super Mario Bros. U and New Super Luigi U that does not normally appear on top of the tower in the world map. He does, however, briefly appear on top of the tower after his defeat from the player or if the player is defeated and returns to the world map.
Mario Golf: World Tour
In Mario Golf: World Tour, a Boss Sumo Bro costume is available for Miis to wear, and they can also use a ball and set of golf clubs designed based on a Boss Sumo Bro.
Minecraft
In the Super Mario Mash-up in Minecraft, Piglin Brutes are replaced by Boss Sumo Bros.
Paper Mario: The Origami King
The Boss Sumo Bro appears as a boss in Paper Mario: The Origami King, having been folded into origami by King Olly.
When Mario, Olivia, and Kamek try to enter Bowser's Castle, a thundercloud with the Boss Sumo Bro on top appears and nearly strikes the former two with lightning, closing the path. Bowser Jr. attempts to get to the cloud in his Junior Clown Car, but is cut up by Scissors before he can reach it. If Mario attempts to walk under Boss Sumo Bro's cloud after this, Boss Sumo Bro strikes him with lightning and deals 36 damage. Once Bowser Jr. is restored to normal, the group uses the Junior Clown Car to get onto the cloud, and Mario and Olivia battle them.
When the battle starts, Boss Sumo Bro has moved to a smaller thundercloud, and six regular Sumo Bros appear also on thunderclouds. Four of them will dismount and steal ON, +1, Fire Vellumental, and Attack icons from the battle rings, and taking them to the center. The other two Sumo Bros. will then use Thunder Guard, electrifying the two spaces in front of them; if Mario steps on an electrified space, he is dealt 6 damage and wastes his turn. Mario can attack the Sumo Bros. on foot by ending up in front of them, and must get back the icon they are holding by defeating them. If Mario is not in front of a Sumo Bro, Jump attacks will hit the Boss Sumo Bro and Hammer attacks will do nothing. If a Vellumental is used, it will hit only the enemies on foot; the Fire Vellumental deals 200 damage, instantly defeating such enemies, while the Earth Vellumental deals 100 damage, enough to instantly KO the normal Sumo Bros., but not the Boss Sumo Bro.
As with all spiked enemies, Boss Sumo Bro and his minions cannot be jumped on (except with Iron Boots), but he can be hit with anything else without an issue.
When the enemies attack, the Boss Sumo Bro and Sumo Bros. in the air use Thunder, the Sumo Bros. dealing 10-14 damage each, and the Boss Sumo Bro dealing 26-28 damage. However, if Mario has used the Earth Vellumental prior, he will completely avoid the attacks due to the Sumo Bros. not being high enough to hit him.
After all the Sumo Bros. on foot are defeated, the remaining enemies will dismount, the Sumo Bros. stealing both Vellumental spaces, and the Boss Sumo Bro charging up Heavy Thunder every turn, which deals 40 damage, but can also be avoided using the Earth Vellumental.

In Super Mario Party Jamboree, Boss Sumo Bro appears as the boss of Boss Sumo Bro Blitzers. Players have to reassemble a broken circuit in order to damage them. If a player fails to reassemble the circuit, Boss Sumo Bro will attack them.
