The Flopside Pit of 100 Trials is a 100-room labyrinth in Super Paper Mario just like in Paper Mario: The Thousand-Year Door. However in this game there are two pits – one in Flopside and one in Flipside called the Flipside Pit of 100 Trials. Both were created to test the forces that guard the Pure Hearts. The Flopside version was used to study the powers of darkness. Accessing the Flopside Pit is done in a manner similar to its Flipside counterpart, except the switch that opens the entrance does not appear until the Flipside Pit is cleared.

In the Pit a random enemy in each room holds a key to a locked door that proceeds to the next room. Every 10th room, a valuable item awaits. Shadoo is the boss of the pit. The pit is designed to look like the LCD screen of an old Game & Watch System.

Both pits have artwork on the walls depicting a Pure Heart and where it is found, which changes every 25 rooms. For example, in the Flopside pit, Rooms 26-50 have a Pure Heart paired with King Croacus I and Floro Sapiens (a reference to Mario's sixth Pure Heart).

The enemies in the Flopside Pit are far more dangerous than their Flipside counterparts, as they are all Dark versions, which are found nowhere else it is thought that Shadoo; the monster at the bottom of the same pit created it with it's power to clone anyone, a technique similar to Doopliss from the previous paper Mario game in reverse. It is recommended that first-time players avoid the Pit until the climax of the game at the very least, with Mario at a high level. By that point, the player should have enough HP and attack power to deal with the enemies. As with the Flipside Pit, stocking up on healing items is strongly encouraged such as the legendary recipe, Dyllis Deluxe made by of course Dyllis.

The first time Mario's group makes it to the 100th room, they will be sent back to the entrance, and will have to make their way through the pit a second time in order to fight the boss.
