Similar to previous titles, most of the bosses in ''Super Mario Galaxy'' are stronger counterparts to normal enemies, and all except the [[Mandibug Stack]] have the background music change when encountered. Most tactics to defeat bosses are subtly conveyed in the level design before they are reached, giving the player an understanding of what to do to defeat them. For instance, prior to facing Dino Piranha in the Good Egg Galaxy, a player taking the galaxy's main path will encounter a [[Spiny Piranha Plant]] surrounded by [[rubbery bulb]]s that can be used to damage and defeat it, which is echoed with how Dino Piranha's clubbed tail must be swung against to damage the boss. Some of these examples are even done between multiple missions and in different galaxies. Kingfin's "tutorial," for example, is in the [[Drip Drop Galaxy]] rather than the [[Bonefin Galaxy]] where he is encountered, and to ensure the player has completed said instructional mission, the Bonefin Galaxy does not become available until it is completed.
Most bosses take three hits to their weak point to defeat, like in most previous games. The Mandibug Stack only takes two, however, and both Megaleg and Mecha-Bowser instead must be climbed and destroyed with a certain attack at the top. Most bosses become faster and more offensive the more hits they have taken, which is often reflected with them becoming visibly enraged and sometimes even glowing red. Some even switch tactics completely midway through the fight, such as Bugaboom taking flight or Bouldergeist spawning a massive pair of hands. Some will also spawn minions the further their battle goes.
Some bosses are fought more than once, and if fought again in a later galaxy will have additional tactics and mechanics to make the battle harder. Ones with especially apparent differences to their fights even get a color change and are listed as separate bosses in official paratext, despite sharing a name and a base model. Otherwise, most rematches are done with the added rules from a [[Prankster Comet]], specifically a [[Prankster Comet#Speedy Comet|Speedy Comet]] or a [[Prankster Comet#Daredevil Comet|Daredevil Comet]].
