Spike's Gambit is divided into three levels of difficulty. The first level takes play during day, with more limited coins and spiked rollers than later levels. Level two takes place during the evening; [[Red Coin]]s, which give three coins at once, start to appear, and after some time has passed, the three Spikes throw a massive spiked roller. This spiked roller can only be avoided by hiding in the alcoves, for it covers the entire horizontal length of the sand. Hitting it causes the player to tumble a great distance down the hill, though coins touched during this tumble are still collected. The third level takes place at night, following the same rules as level two but with even more coins and spiked rollers.

In Koopathon mode, Spike's Gambit is played whilst competing against other players, one level at a time. In Free Play mode, the minigame is played individually, with all three levels played in a row with items limited to the Coin Bag and [[Double Medal]]. The objective of the Free Play variant is to simply obtain the most amount of coins in a single game. The player's high score, if excessing either the current score or the default score of 30 coins, is recorded, and can be compared with other top player's score online in the Party Plaza. Obtaining at least 350 coins in a single game in Free Play mode unlocks the "Spike Seeker" achievement.
