The opening cinematic starts with a countdown, as the characters prepare their golf attire and equipment before they enter the Super Golf Stadium. All characters start to swing their golf clubs, with Mario swinging his to strike the golf ball. After taking their first shots, they run towards their golf balls, with Waluigi and Luigi pushing each other around and Mario and Bowser looking at each other.
 
Daisy uses her Special Dash to leap over Peach. Peach responds with her [[Special Shot (Mario Golf: Super Rush)|Special Shot]] that sends the golf ball flying. Wario and Waluigi spot Mario and Bowser, and Wario uses his Special Dash while Bowser breathes fire, but Mario leaps over both. Mario scores his first point by getting the golf ball into a hole. As the game continues, Mario and Bowser are in fierce competition.  
 
A Chain Chomp appears from one hole and chases Wario, who runs into a pillar. He then spots a Banana item, which upon using it trips up Bowser and Waluigi. Mario uses his Special Shot, but because Waluigi bumped into a Switch block, the ball ricochets and lands on King Bob-omb, who rolls downhill and explodes on Bowser, Wario, and Waluigi. The explosion also rolls Mario's golf ball into a hole to give him the last point to win the game. While Peach and Luigi cheer him on, Mario is confused by the events that happened, but gives a thumbs up pose. Meanwhile, Bowser, Wario, and Waluigi are grumbling from their defeat, when Bowser spews black smoke, leading to the title screen.

Chain Chomps charge at any player within their vicinity to damage them; Thwomps smash the spot that they are on at intervals, causing anything under them to be blown away; Whomps act as walls that block the ball. Two items that appear in the courses are [[Bob-omb]]s and [[Banana]]s. When a player go up to one of those item, they will aim them at the player with the highest placement. The Bob-ombs will cause an explosion to occur within the radius, while the Bananas will be split into eight and can cause the players to slip. Damaged players will, in addition to being momentarily stunned, drop coins and lose some of the Special Shot gauge. The coins can be used to fuel their Special Shot gauge. Both the Whomps and the Bananas only appear in the Technical course. The Mii is not initially playable in this mode, and can only be unlocked for use by completing the story mode.

This mode is added in version 4.0.0. In '''Target Golf''', there are three targets that are lit up, each consisting of four scoring zones with a flag in the middle (there are no holes to sink the golf ball in). In this mode, players take turns to hit from one of the multiple different tees to one of the targets. In order to obtain points, the player must land the golf ball on one of the zones, which will grant points to the player based on how close it is to the flag. From the outermost zone to the innermost zone, the points granted are: 10 points, 20 points, 30 points, and 40 points. After points are obtained from landing the golf ball on that part of the zone, it stops lighting up and is considered knocked out, meaning that landing on the same part of the zone won't grant points to another player. When all of a zone's circles have been knocked out, the last player who caused it gets a Dead-Aim Bonus, which grants an additional 30 points on top of the points granted, although this bonus is not affected by any multiplier. On certain shots, which is during the turn of the player with the least amount of points in multiplayer and the last of every five shots in Solo Challenge, a star target will light up, which has three zones. The amount of points granted from landing the ball within the outermost zone to the innermost zone are: 20 points, 40 points, and 100 points. After the golf ball lands on any part of this star target, the rest of the zones stop lighting up.
 
The club set determines the types of clubs available. The given options are the same as in Speed Golf, with the difference that the putter is not available as an option. Three different courses are available in target golf: Standard, Forest, and Volcano. These courses are different from any other course in the game, even the ones in Battle Golf. The Standard course has a simple layout with no obstacles and few elevation changes, the Forest course has a woodsy layout featuring trees, a river, and longer distances from tees to targets, and the Volcano course has a tricky layout featuring lava and some elevation changes. In every course, there are four different tee points, offering different vantage points from the targets. Tee changes happen differently between multiplayer and Solo Challenge. In multiplayer, they happen after everybody has taken a shot, while in Solo Challenge, they happen after every five shots are taken.  


Whenever all parts of one target are knocked out, it will only be reset after every player took their turn. However, if all the targets stop lighting up before at least one player gets to tee off within that turn, one of the targets will be reset before everybody's turn ends, to give a chance for them to score. In Solo Challenge's Target Golf, the board reset works differently. Instead of the target resetting immediately after all the zones stop lighting up, fully knocked out targets will only reset after every fifth shot.

This mode is added in version 2.0.0. In '''Ranked Match''', the player can participate in matches with fixed settings. Once selected, there are four different categories with separate rankings: "Button Controls: Standard Golf", "Button Controls: Speed Golf", "Motion Controls: Standard Golf", and "Motion Controls: Speed Golf". Every match is participated by four players, and if the amount of players did not reach that amount after the timer reaches zero, CPU players at C- rank will fill in the spots. If two or more players start a match, there will be a 10-second timer before the match is initiated. The rank and amount of points for each category is recorded each month, and when the month is over, the records can be viewed within Ranked Match Records.


The golf courses selected for every category is different, and they all change courses to the next one in the course order list after every 30 minutes. For New Donk City and Shelltop Sanctuary, their Pro courses layouts are used in ranked matches. Some settings are shared between every category, namely that 3 holes are played per match, Mii characters are prohibited, and Tee Handicaps are enabled except for Rookie Course, New Donk City, and Shelltop Sanctuary. Tee Handicaps basically indicate the tee that the player start from based on their ranking, where they will start further away the higher their ranking is. In Standard Golf, players get to tee off at the same time as opposed to taking turns, while in Speed Golf, every player starts at the same time regardless of how well they did in the previous hole. If one of the players completed a hole, there will be a 60 second limit for every other player to complete the hole, otherwise it will be an automatic forfeit. One difference in setting between the modes is that "Button Controls: Speed Golf" is the only one that allows Special Shots while the others disallows it, though "Button Controls: Standard Golf" allowed it prior to version 3.0.0. A difference between Standard Golf and Speed Golf in both control methods is that the former's scoring format is Stroke Play (score is based on how well below par the player is), while the latter's scoring format is High Score (score is based on the rank each hole is finished).  
In Battle Golf, four players compete in a stadium filled with nine holes and a number of obstacles to be the first player to sink three holes.<ref name="Overview"/> In the stadium, players can collect coins and can even launch [[Bob-omb]]s with their golf club to damage their opponents.


After three holes are completed, the player is given points based on how well they did. These points will determine the rank the player falls under, where they will start from zero points every month. For Standard Golf, the amount of points given is 2 points for a score of +3, with an extra 2 points for every stroke reduced from that amount, while in Speed Golf it's based on the amount of points earned in that mode. For Participation Bonus, 3 points are granted if the player finished one hole, but will be zero if they forfeit every one of the three holes since version 3.0.0. For Higher-Rank Defeat Bonus, if the player obtained a higher score compared to a player with a higher rank, 10 points will be granted for each opponent the player has a better score than, though it will not be granted if the player defeated another with a higher sub-rank but still within the same rank.
