The Gahaha Virus is a germ created by Bowser, under the guise of Gahaha Daiō, in order to take over the Mushroom Kingdom in Dr. Mario. The germ infect the people by making them laugh without rest, eventually leading to death (which, in the evolved state, happens after 6 hours): the Gahaha Biyō. The germs look like normal amebas with arm, legs, and a mischievous face.
During the Ikebana Expo in the Mushroom Kingdom, Gahaha Daiō sneaks in and released three Dokudoku Pakkun Flower that can spit the Gahaha Virus. Soon enough, everyone in the Kingdom is infected, except for Mario who quickly covers his mouth and nose. With his medical kit, the self-proclaimed Dr. Mario studies one of the Virus he managed to trap in a Petri Dish, discovering that it is immune to any known cure. After noticing it that one mountain region of the Kingdom was uninfected, Mario, alongside Princess Peach and Princess Daisy (who were the first to be infected), flies to the mountains, where he is awaited by Gahaha Daiō. He gives Mario an apple filled with ten doses of Gahaha Viruses, but the princesses eat it instead and start to have a seizure from laughing. Gahaha Daiō then try to infect the village that escaped the infection, but one mushroom repels both the Viruses and the Dokudoku Pakkun Flower, the Miracle Kinoko. After giving a bite of it to the princesses, curing them and saving them at the last second, Dr. Mario creates medicinal capsules containing pieces of the Miracle Kinoko to kill the Plants and to cure the Kingdom from the Virus.
However, one Gahaha Virus remained, the one in Mario's Petri dish, and after reuniting with Gahaha Daiō, the latter transforms the germ in the Red, Blue, and Yellow Viruses. In doing so, Bowser has decomposed their weakness to the Miracle Kinoko, making them immune to it. The three Viruses then sneaks inside Mario's house and enter in Luigi's body, infecting him, by not only making him laugh, but by also generating a wall of Gahaha Viruses (immune like them) in his stomach and his brain, which will lead to his death in 6 hours. In this form, however, the Gahaha Viruses have one weakness: if one Virus is aligned with at least three elements of the same color, it will disappear. Using this, Dr. Mario reworks the capsules and vanquishes the Viruses from his brother's body.
In the last chapter, Gahaha Daiō tries one more time to beat Dr. Mario by challenging him to a VS. game of Dr. Mario, once again using the Gahaha Viruses as the Viruses to clear form their giant beakers, but does also threats the crowd spectating the match to infect them with regular Gahaha Viruses by releasing them from a party ball if Mario loses the match. Dr. Mario ends up winning and, before Gahaha Daiō or his three Viruses can open it.
