Doopliss later helps Beldam and Marilyn in an attempt to prevent Mario and his team from reaching [[Poshley Heights]] by the [[Excess Express]]. Disguised as the star [[Zip Toad]], he steals [[Nitro Honey Syrup]] from the [[Businessman]], [[Shell Earrings]] from the the [[Excess Express waitress]], and [[Toodles]]'s [[Gold Ring (item)|Gold Ring]]. Beldam and Doopliss plan to create a "[[Threatening Note|sticky, yummy, gooey mess]]" by combining the syrup, calcium from the earrings, and gold to create a bomb which would blow up the train. Doopliss Zip Toad is stopped at the last minute by Mario. When [[Pennington]] arrests Zip and takes him off the train at [[Riverside Station]], he shocks Pennington by turning back and dashing away. Doopliss, Beldam, and Marilyn reappear in Poshley Heights, where they break into the [[Poshley Sanctum]] by smashing a window and steal the [[Garnet Star]], which turns out to be a red herring created by Pennington, while Mario and his team find the real one hidden inside the painting.

Here, Doopliss, Beldam, and Marilyn fight Mario and his team in front of the [[Palace of Shadow Tower]]. In this battle, Doopliss is a bit tougher, and has the ability to transform into Mario or one of his partners, and attacking as them. Goombella's field [[tattle]] for Doopliss is viewable only after he, Beldam, and Marilyn are defeated. Following their defeat, Doopliss, Beldam, and Marilyn watch Mario's battle against the [[Shadow Queen]]. After the Shadow Queen is defeated, Doopliss wants everyone to call him by his real name, tired of Beldam calling him a "freak-in-a-sheet".

This special syrup supposedly causes an enormous explosion when mixed with calcium and gold, so he prepares its use as a bomb, stealing the [[Shell Earrings]] of the [[Toad Waitress]] for the calcium part and [[Toodles]]'s [[Gold Ring]] for the gold part, but is stopped in last minute by Mario. When [[Pennington]] arrests Zip, and takes him off the train at [[Riverside Station]], he shocks Pennington by turning back and dashing away.  


In the Nintendo Switch remake's ending credits, Doopliss appears alongside the other Chapter 4 characters. He walks along the stage accompanied by a gang of [[Boo]]s. He then transforms into his Mario disguise and tips his hat to the [[audience]] before he and the Boos leave.

Doopliss also makes an appearance in the manga ''[[Super Mario-kun]]'' in chapter 9 of [[Super Mario-kun Volume 33|volume 33]]. Doopliss only plays the Twilight Town boss role in the manga, while his alliance with Beldam and Marilyn is not depicted. Under the request of the cursed Twilighters, Mario, Goombella, and Koops (who was cursed like the other Twilighters and is now half-pig) face Doopliss, who is using the Ruby Star to cast his curse, as he shows by removing the curse on Koops. Mario goes for a punch and Doopliss immediately surrenders as he does not like confrontation, which is also why he cast the curse, to take over the village by turning the inhabitants into harmless pigs. As Mario then reaches for the Crystal Star, he trips onto Doopliss, headbonking him and swapping bodies with him. Doopliss tries to take advantage of the situation, but Mario stops him and tries to reason with Koops, but he responds by hitting him with his shell. After a while, Goombella realizes the trick and shoves Doopliss onto Mario, headbonking them and swapping their bodies back to normal. Doopliss is then forgiven and promises to become a stronger and better person to be accepted by the Twilighters, which are turned to their normal selves by Mario.


Alongside the other friends of the adventures, Doopliss is then seen in chapter 5 of [[Super Mario-kun volume 35|volume 35]] rooting for Mario and the team via the Crystal Stars during the final battle.

In the GameCube version of the game, if Mario is recolored from equipping either the [[W Emblem]] or [[L Emblem]] badges, Doopliss as Mario will still have Mario's normal clothes. Doopliss enchanted the Creepy Steeple's bell to turn an inhabitant of Twilight Town into a [[pig]] each time it rings. As explained by Vivian, since nobody knows his name, he gains the ability to cast incredible magic (a common theme in real mythology), which is likely amplified by the Crystal Star he possesses. This power is what allowed him to tie his Duplighost abilities to the Steeple's bell. During his boss theme, the bell rings fully four times in every loop. Also, the bell might still be enchanted, as it is stated that it no longer rings, though it is most likely that it is not.
Doopliss somehow enchants the Creepy Steeple's bell to turn an inhabitant of Twilight Town into a [[pig]] (similar to a [[lil' oink]]) each time it rings. It is unknown how exactly he does it, and it is also never revealed how the curse is lifted. One possibility is that Doopliss' defeat brought the curse down. A similar situation occurs in the second fight, where he is revived with a Life Mushroom so that the fight cannot be completed until [[Vivian]] rejoins the fight.
