Power-up Panels is a minigame that is played in Red Toad Houses in New Super Mario Bros. Wii. It takes place in a room with a six-by-three grid of Flip Panels decorated with question marks. The back side of each has an image of a power-up, Bowser, or Bowser Jr. Players must take turns climbing around the fence and punching the panels, causing them to flip and reveal their backsides. If two Bowser images or two Bowser Jr. images are matched, the game ends. If all item panels are revealed, the game ends and the characters will rejoice. At the end of the game, players receive one item for every pair of identical panels with revealed faces.

- Super Mushroom (World 1)
- Penguin Suit (World 3 to 9)
- Mini Mushroom (World 2 to 9)
- Super Star (has a 1/3 chance of appearing in each world)

Trivia
- When matching two Bowser or Bowser Jr. panels, the small tune from Super Mario 64 when trying to open a door in the castle without enough Stars or a key is heard.
- If the player gets a perfect score, the fanfare when returning to the castle after obtaining a Power Star also from Super Mario 64 is heard.
- This minigame is similar to the N-Mark Spade Panel from Super Mario Bros. 3.
- If playing with two or more players, it is possible to steal another player's turn by Ground Pounding on them during their turn.
