Endurance Alley is a minigame mode in Mario Party 6 hosted by Twila in the Minigame Mode. To unlock Endurance Alley, the player must have unlocked at least one 4-player minigame, one 1-vs-3 minigame, and one Duel Minigame (Bonus Minigames and Mic-Minigames do not count).
In Endurance Alley, the players walk through a dark alley while playing a series of minigames. The objective is to get 100 consecutive wins. The game ends when players lose one of the minigames that they must play.

Trivia
- It is possible to complete Endurance Alley by playing only three minigames (one 4-player, one 1 vs. 3, and one duel) 100 times in a row.
- In the PAL version, Same Is Lame, Trap Ease Artist, Pitifall, and Trick or Tree are not played in Endurance Alley, most likely due to those mini-games' reliance on luck.
- The music that plays for the first 30 minigames is a remix of the Super Mario Bros. underground theme. This remix would itself be remixed in Mario Party 7's King of the River.
