If there is an item nearby (ex. Banana) and Donkey Kong claps, he will reach for the item and grab it. If there is an enemy nearby and Donkey Kong claps, the enemy might be defeated.

Clapping has many other uses in the game. For example, in some boss battles, clapping causes Donkey Kong to avoid certain attacks.

Several clap moves appear as Donkey Kong's Smash Attacks, most notably his side Smash and up Smash, in Super Smash Bros. series.
